Description:
Mini-Prep 24 and Mini-Prep 48 benchtop instruments automatically purify plasmid DNA directly from bacterial culture. In just over an hour, both produce high-purity plasmid DNA that can be used in most molecular biology processes, including manual and fluorescent automated DNA sequencing. Cuts to completion with restriction enzymes. No centrifugation step is required; allows for direct loading and processing of up to 24 crude bacterial cultures.

Internal electrophoresis processing rig supports precast disposable sample cassettes; cassette lanes have microtiter-plate spacing for use with multichannel pipetters. Each cassette processes up to twelve 2.2mL bacterial culture samples. Mini-Prep 24 processes up to 24 minipreps per hour; Mini-Prep 48, 48 minipreps per hour. Yields of 4 to 8µg of high-copy-number plasmids may be produced from 2mL culture. For best results, use with High-Yield Bacterial Growth Media (MR2001-3, -10, and -20).

Four efficient, automatic steps. Bacterial lysis occurs directly in the culture media in cassette wells separated by plastic partitions. Enzymes and reagents that dissolve bacterial membranes and degrade RNA and proteins are contained in the tablets and applicator comb supplied with each cassette.

The instrument automatically fills with running buffer; a microprocessor-controlled electrophoretic process then separates the plasmid DNA from the digested cellular components. During the separation process, plasmid DNA ranging in size from 2 to 20kb is electrophoresed until the 2kb supercoiled plasmids run to the end of the separation gel.

Purified plasmid samples are electroeluted and concentrated in the microchamber block, which is automatically pushed into place. The microchamber block contains a high cut-off filtration membrane that collects the purified DNA as a film on its inner vertical surface.

After the electroelution stage, the running buffer automatically drains. Purified DNA collects in a 25µL droplet in the bottom corner of the microchamber block window; it can be removed with a disposable tip pipetter or a multichannel pipetter with microtiter-plate spacing
